PPTP Acute Lymphoblastic Leukemia Xenograft Testing Laboratory

This laboratory employs multiple xenograft models of human acute lymphoblastic leukemia (ALL) that have been directly established in nod/SCID mice. These xenografts have been established from patients with relapse/refractory ALL and also from patients who had responsive disease.

ALL cells are injected into mice via the tail vein and engraftment of the ALL cells determined by monitoring circulating ALL cells in the blood using flow cytometry and anti- human CD45 (1, 2).

Clinical Trials Enabled by this Laboratory

Data generated by the PPTP screen showing activity of bortezomib in the ALL xenografts confirmed data obtained in vitro by other investigators and helped enable the ongoing TACL phase I study of bortezomib + vincristine, dexamethasone, and L-aspariginase (T2005-003).
